|---|---|---|
| PI | principal investigator,
| RA | research assistant,
It didn't work
Dr. Okafor(生物系副教授,实验室有六个人,回邮件很快) You(大二,想找研究机会,之前发过两封冷邮件没回音)
Dr. Okafor: Come in. You’re the one who emailed about the microbiome project?
You: Yes. Thanks for replying — I know you get a lot of these.
Dr. Okafor: I do. Yours was four sentences and it had a specific question, so I answered it. What’s your background?
You: I’m a sophomore in biology. I’ve taken the intro sequence and organic chem. I did the lab sections, but nothing beyond class.
Dr. Okafor: Have you done any coding?
💬 Okafor
You: A little Python. I analyzed my own data for a class project.
Dr. Okafor: That’s more useful than you think. Half of what we do now is analysis, not pipetting.
You: That’s actually why I emailed. I want to learn the analysis side.
Dr. Okafor: Okay. Let me be straight with you about what this looks like.
💬 Okafor
You: Please.
Dr. Okafor: First semester, you’ll do ten hours a week and most of it will be boring. Labeling tubes. Running the same protocol forty times.
You: I expected that. I’d rather learn it properly than start in the middle.
Dr. Okafor: Good answer. Do you want credit or pay?
💬 Okafor
You: What’s the difference?
Dr. Okafor: Credit means you register for research units and write a report. Pay means I need funding, and I have one slot.
You: Which do you recommend for a first semester?
Dr. Okafor: Credit. It’s easier for me, and you get a transcript line. If it goes well, we talk about pay in the summer.
You: Credit works for me. How do I register?
Dr. Okafor: There’s a form the department needs, and you’ll have to complete safety training before you touch anything.
You: How long does the training take?
Dr. Okafor: Two hours online, then a walkthrough with Hana. She runs the day-to-day.
You: Hana’s the PhD student?
Dr. Okafor: Third year. She’ll be your first stop for everything. Ask her before you ask me.
You: Understood. One more thing — how many hours a week in the summer?
Dr. Okafor: Full time. Forty hours, and that’s when you’d actually get a project of your own.
You: That’s what I was hoping. Can I start this week?
Dr. Okafor: Finish the training, email me the certificate, and Hana will set up a time. And one rule — if you’re going to miss a shift, tell Hana, not me.
You: Got it. Thanks for the chance.
Dr. Okafor: Don’t thank me yet. Thank me in November when you still show up.

Dear Professor, I am a student who is very interested in your research. | 空话开头,教授看不到重点 | I read your 2024 paper on gut microbiome stability — could I ask about the sampling method? |
I want to learn something in your lab. | 太笼统,且以"我要"为中心 | I'd like to learn the analysis side. I have one slot free this semester. |
Please give me a chance, I will work very hard. | 求机会,不如给证据 | I've taken organic chem and I can write basic Python. |
I am free anytime you need me. | 没有边界,反而不可靠 | I can do Tuesday and Thursday afternoons, ten hours a week. |
Sorry to bother you, I know you are very busy. | 铺垫太长,浪费第一句 | Thanks for replying — I know you get a lot of these. |
I hope you can accept me. | 像申请,不像合作 | Would you have a spot for an undergrad this semester? |
Yours had a specific question, so I answered it.
I'd rather learn it properly than start in the middle
3. credit or pay
美国大学实验室有强制的 safety training(通常线上课程 + 现场走查),有些还要求年度更新。
Hana(博士三年级,负责带新人,说话快、直接) You(第一天进实验室,戴着新买的白大褂)
Hana: Okay, tour time. First rule: where do you put your bag?
You: Um … on the bench?
Hana: No. Never on the bench. There’s a hook by the door. Bench space is for experiments only.
Hana: Don’t apologize, just remember. Second: gloves. When do you wear them?
You: When I’m handling chemicals?
Hana: And when you touch anything that touched a sample. And you take them off before you touch the door handle.
Hana: Everybody forgets. That’s why the door handle has a sign on it.
You: Okay. What about the fridge with the red tape?
Hana: That’s the -80. Do not open it unless you know exactly what you’re getting. Every time it opens, it costs us.
Hana: Temperature recovery, and the samples. Some of those go back eight years.
You: Okay, noted. And the log book?
Hana: Every time you take something out, you write it down. Date, your initials, what you took, how much.
You: What if I forget?
Hana: Then nobody knows where it went, and the whole lab stops for a day to figure it out. That’s happened twice.
Hana: It’s not pressure. It’s just a habit. Takes four seconds.
You: Fair. What about mistakes — what if I break something?
Hana: Tell me immediately. Not at the end of the day. Immediately.
You: Even if it’s small?
Hana: Especially if it’s small. Small spills become big problems when nobody mentions them.
You: Okay. And if I don’t know how to do something?
Hana: Ask. I would much rather answer a question than redo an experiment.
You: Good. That’s my main skill right now — not knowing how to do things.
Hana: That’s fine for week one. By week four you should be able to prep the buffer without asking.
You: What should I do today?
Hana: Shadow me. Watch the whole protocol, don’t touch anything, and write down every step.
You: Write it down even though you have a printed protocol?
Hana: Yes. The printed one is the ideal version. Mine has the seven places it goes wrong.
You: That’s the version I want.
Hana: Exactly. Okay, gloves on. We start with the lysis step.

Tell me immediately. Not at the end of the day.
5. I'd rather answer a question than redo an experiment
Dr. Okafor(PI,组会主持) Hana(博士三年级) You(第一次在组会上汇报,做了四周实验)
Dr. Okafor: Okay, next up. Wei, you’re on.
💬 Okafor
You: Hi everyone. Um, so this is my fourth week. I’ve been running the extraction protocol.
Dr. Okafor: How many samples?
💬 Okafor
You: Twenty-four. But the yields were low on most of them.
Dr. Okafor: How low?
💬 Okafor
Dr. Okafor: Okay. What did you try?
💬 Okafor
You: I re-ran six of them. Same result. So I don’t think it’s my technique.
Dr. Okafor: What else could it be?
💬 Okafor
Dr. Okafor: Good. Hana, thoughts?
Hana: Did you check the lysis time? We had a batch of enzyme that was weak last year.
You: I didn’t. How would I check that?
Hana: Run a positive control with the old batch and compare. I have some in the -80.
Hana: Do it Wednesday. I’ll show you the setup.
Dr. Okafor: Anything else on this? Okay. Now — what’s your next step after the control?
You: If the enzyme’s the problem, I switch batches and re-run everything. If it’s not, I look at the tissue source.
Dr. Okafor: Good. That’s a plan. One thing about how you presented this.
You: Yes?
Dr. Okafor: You said “it didn’t work” three times in two minutes. Don’t do that.
You: Sorry — I thought I should be honest.
Dr. Okafor: You should. But lead with what you learned, not with what failed. “Low yields on twenty-one of twenty-four, consistent across repeats” is a finding.
You: Okay. So the consistency is the information.
Dr. Okafor: Exactly. A failure that repeats is data. A failure that doesn’t repeat is noise. Say which one you have.
You: Noted. And — should I send slides before the meeting next time?
Dr. Okafor: Yes. Sunday night. Then people come in already thinking about your problem.
Hana: One more tip. Put the negative result on the first slide.
You: Why the first slide?
Hana: Because that’s the thing you need help with. Don’t make them wait until slide eight.
You: That’s the opposite of what I’d do.
Hana: I know. Everyone does it backwards their first month.

Low yields on twenty-one of twenty-four, consistent across repeats. I don't know why yet. My next step is a positive control. ——
2. It didn't work 说一次就够。
A failure that repeats is data
Sunday night
I don't know yet. That's why I'm bringing it here.
Hana
Hana(博士三年级,正在写论文,桌上三个显示器) You(第三次失败,情绪有点崩)
You: Hana, do you have five minutes? I’m kind of stuck.
Hana: Yeah, sit. What’s going on?
You: Third failure in a row. Same step. I did everything the same way as the first time, and the first time worked.
Hana: Show me your notebook.
You: Here. The last three runs are on this page.
Hana: … Okay. What’s missing here?
You: I don’t see anything missing. That’s the problem.
Hana: The room temperature. You wrote it for the first run and not for the others.
You: Does that matter? It’s a lab, it’s always cold.
Hana: It was eighty-two degrees on Tuesday. The AC was broken. Half the building was complaining.
Hana: Yeah. That step is temperature-sensitive. It’s in the original paper, in a footnote.
You: I read the paper three times and I missed the footnote.
Hana: Everyone misses the footnote. That’s why you write the temperature down every single time.
Hana: You have to throw out the data. The three days taught you what to record. That’s not nothing.
You: It feels like nothing right now.
Hana: I know. I lost four months in my second year. Want to hear about it?
Hana: The incubator was off by two degrees for a whole summer. Nobody noticed until August.
You: What did you do?
Hana: Cried for a day, then started a log on the wall. Every morning, first thing. It’s still there.
You: So the lesson is: log everything.
Hana: The lesson is: log the things that can change. Temperature, batch numbers, who else used the room.
Hana: Start Monday, but first re-read the methods section with a highlighter. Every condition, not just the steps.
Hana: And tell Dr. Okafor at the next meeting. Not because you did something wrong — because the group should know the AC is a variable.
You: I’d rather not advertise three failures.
Hana: That’s the wrong instinct. If someone else runs that step next week, you just saved them three days.
Hana: Go home. You’re not going to fix it tonight.
Hana: Every single person in this lab has felt that. Including him. He told me so.
You: Dr. Okafor?
💬 你: Okafor 教授?
Hana: Second year of his PhD. He almost quit. Now go home.

|---|---|---|
I am so stupid, I wasted three days. | 自责不解决问题,也让对方难接话 | Third failure in a row — same step. Can you look at my notebook? |
Maybe I am not suitable for research. | 结论下得太早,且对方只能安慰你 | Is it normal to feel like you're bad at this? |
I don't want to tell others about my failure. | 隐瞒会让别人重复踩坑 | I'll report it at the next meeting so nobody repeats it. |
Please help me find the reason. | 把排查责任全交出去 | I re-ran it three times with the same setup — what should I check? |
I will work harder next time. | 努力不是变量 | I'll log temperature and batch number every run. |
The experiment is too difficult for me. | 归因错误,且没有信息 | The step is temperature-sensitive and I didn't control for it. |
If someone else runs that step next week, you just saved them three days. ——
4. Is it normal to feel like you're bad at this?
You're not going to fix it tonight. ——
Dr. Okafor(PI,暑期项目结束前两周) You(做了十周全职,有了一些结果)
Dr. Okafor: Sit down. I looked at your summer data. This is real.
You: Real how? I mean — is it publishable?
Dr. Okafor: The main result, yes. It needs two more experiments, and then we write it up.
You: I’d like to be involved in the writing. Even if it’s just editing.
Dr. Okafor: Good. You should be. Now — let’s talk about authorship, because I want to do this before the paper exists, not after.
You: Okay. What’s the convention here?
Dr. Okafor: First author is whoever did the work and wrote the draft. That’s you, if you finish the two experiments.
You: And you’d be last?
Dr. Okafor: Last, and corresponding author. Hana would be second — she designed the assay you’re using.
You: That seems fair. What if I can’t finish the two experiments before school starts?
Dr. Okafor: Then we talk about it again. Authorship follows contribution, and contribution can change.
You: Is that in writing anywhere? I’ve heard stories about people getting dropped.
Dr. Okafor: It’s not a contract, but I’ll put it in an email to you and Hana. That’s how it’s usually handled.
Dr. Okafor: Have you thought about what you want to do next? Grad school?
💬 Okafor
You: I’m thinking about it. But I’m not sure I want a PhD.
Dr. Okafor: Good. Don’t do one because you liked one summer.
💬 Okafor
You: What should I do instead?
Dr. Okafor: Do a second summer somewhere else. Different lab, different field, different people. Then decide.
You: That’s not what I expected a professor to say.
Dr. Okafor: I get paid either way. You’re the one who spends five years.
💬 Okafor
You: Fair. Can I ask you for a recommendation letter?
Dr. Okafor: Yes. When do you need it?
💬 Okafor
You: The first deadline is December 1st.
Dr. Okafor: Then send me everything by November 1st. Resume, your statement, the list of programs, and the deadlines.
You: A month ahead. Noted.
Dr. Okafor: And ask in person, like you just did. Email me the materials, but don’t ask by email.
You: Why does that matter?
Dr. Okafor: It means you thought about who you’re asking. It takes thirty seconds and it changes how I write it.
You: Okay. Thank you — for the summer, and for saying the authorship thing out loud.
Dr. Okafor: Everyone deserves to know the rules before they play.
💬 Okafor

|---|---|---|
Can I be the first author? | 直接要位置,太硬 | What's the authorship convention here? |
I worked very hard this summer. | 努力不等于贡献,且听起来在讨价 | I designed the third experiment and wrote the analysis code. |
Please write me a recommendation letter. | 邮件里这么问很失礼 | Would you be willing to write me a letter? Here's my timeline. |
I need the letter next week. | 时间太赶,教授会拒绝或写得很敷衍 | The first deadline is December 1st — I'll send everything by November 1st. |
I want to go to graduate school because I like research. | 太笼统,且导师会追问 | I'm considering it, but I want a second summer in a different lab first. |
Thank you for your guidance, I learned a lot. | 空洞的感谢 | Thank you for saying the authorship rule out loud. |
Let's talk about authorship before the paper exists, not after —— 论文一旦开始写,各方期待就会固化,这时候再谈容易伤感情。
2. first author 和 corresponding author 是两件事。
I'll put it in an email to you and Hana —— 美国学术圈的习惯做法:重要约定用邮件确认。 (Just to confirm what we discussed…),这不是不信任,是常规操作。

1. Wei, you're on.
2. How low?
3. I don't know yet.
4. Show me your notebook.
5. Don't thank me yet.
